Mr. John's car / Car of  Mr.John
In above given both sentences have the same meaning. 
In first sentence the importance has been given to subject Mr.john while in second sentence the importance has been given to Car .
Example sentences on Apostrophe s ('s) :-

    1. My brother's house./ The house of my brother. 
    2. My heart's feeling. / The feeling of my heart.
    3. My Mobile's cover/ The cover of my mobile.
    4. My father's friend's son./ The son of my father's friend.
    5. My brother's car. / The car of my brother 
    6. Your mobile's volume. / The volume of your mobile. 
    7. Lion's roar / Roar of the lion 
    8. My Teacher's voice / Voice of my teacher.
    9. Football team's player / Player of Football team's.
    10. Girl's brother / Brother of the girl .


Whenever we need to use apostrophe s ('s) after a plural word ending with s, then we must not use "s" after apostrophe.

Example-  These are my brothers' clothes.
                  My friends' dresses are new.
